脂质体包裹血红蛋白的研制及检测(7200字)
摘 要:目的 探讨用猪血红细胞经冻融破膜释放出血红蛋白(Hb),将Hb经过离心、超滤并制成犬血液代用品的新途径。方法 采用脂质体包裹Hb的方法,并将制备脂质体包裹血红蛋白(LEH)通过家兔耳缘静脉换血试验输入体内,异侧耳动脉放等体积血,检测受试家兔血液中的血红蛋白、红细胞、白细胞及血小板。结果 制得Hb平均浓度为141g/L(5批)、纯度为97.7%稳定的LEH,LEH包裹率为27.7%,冰冻保存泄漏率<10%,LEH的直径为(0.85±0.13)μm。结论 LEH是具有生物学活性的新型载氧制剂。

Preparison and Detection on Liposomes—Encapsulated Hemoglobin
Abstract:Objective The hemoglobin (Hb)was made as blood substitute of dogs by freeze-thaw broken film releases hemoglobin(Hb), centrifugal,ultrafiltration ,so the new to make blood substitute of dog was tried .Methods The hemoglobin(Hb)was package by liposin and made into LEH.The LEH was transfused into the rabbit through ears rim vein when it’s blood was input through ears artery.The hemoglobin,Red blood cells,leucocyte and platelets in rabbit blood were detected.The results Hb average concentration was 141g/L (5 batch )、Purity of 97.7%, LEH package rate for 27.7%,Froze preserving the leakage rate<10%,the diameter of LEH for(0.85±0.13)μm..Conclusion The biology active LEH is a new type of carrying oxygen preparations.
